2-cell-like cells (2CLCs) that resemble 2-cell stage embryos can arise with a very low frequency in ES cell cultures. Here, we found a novel factor, Usp7 which promotes 2CLC transition from ESCs and its effect is dependent of DNA replication. Endogenous retroviruses and 2-cell stage specific genes are significantly upregulated in siUSP7-transfected 2CLCs. RNA-seq of siControl and siUSP7 transfected cells. GFP+ are 2CLCs.
