19-36 nt long ncRNA sequences from total RNA of HeLa and HEK-293 cells

HeLa and HEK-293 cells were cultured in DMEM media with 10% FCS at 37 deg. Celsius and cells were harvested at 90% of confluency. Total RNA was isolated from the cells using TRIZOL reagent and small RNA were concentrated using mirVANA microRNa isolation kit. These small RNAs were subject to sequencing on Illumina Genome Analyzer IIx platform using Library Source: Total RNA, Library Selection: Gel selection, ~140 bases(120 are adapter sequences and 20-22 bases are actual insert size) and Library Strategy: Adapter ligation 5' & 3' end of RNA, RT_PCR amplification, gel selection and further QC on bioanalyzer.
