Dendritic cell Differentiation: A case of rapid transcriptome change and unreliable data normalization

The present study evaluated the expression of eleven commonly used housekeeping during the differentiation of bone marrow derived dendritic cells of mice from the precursor cells. The expression of these genes was studied over the period of seven days during the differentiation process. The PCR based expression data of all the genes was also analyzed by gNorm to determine the most stable reference gene. Based upon the M value V value data of gNorm; minimum two reference genes with lowest M value are sufficient for data normalization in current study.

本研究针对小鼠前体细胞分化为骨髓源树突状细胞(bone marrow derived dendritic cells)的过程,评估了11种常用管家基因(housekeeping genes)的表达水平。在整个分化过程的7天周期内,我们对这些基因的表达情况进行了动态监测。本研究还通过gNorm软件对所有基因的基于PCR技术的表达数据进行分析,以筛选出最稳定的内参基因。基于gNorm分析得到的M值与V值数据,本研究中仅需选取M值最低的至少2个内参基因即可满足数据标准化的需求。
