To determine whether the degradation of GFP-RNF152 depends on VPS4 AAA-ATPase. Conclusion: Overexpression of the dominant-negative mutant of VPS4A (E228Q) leads to the accumulation of GFP-RNF152. Notes: The expression of VPS4A WT and E228 is driven by a Tet-on promoter. The expression is induced by adding 2ug/ml of doxycycline for 24 or 48 hours. GFP-RNF152 is stably expressed in the cell.
