The microRNA cluster miR-106a~363 is expressed from the single X chromosome in male mouse ES cells. The cluster has been targeted by homologous recombination and conversion to conditional alleles. I wish to assess the effect of the targeted deletion and reconstitution of the microRNA loci primarily on the targeted loci themselves. For instance whether the supposed nulls are properly null, and whether reconstitution of the miRNA locus has restored expression correctly. The total RNA has been prepared using a Qiagen miRNeasy kit. The RNA samples available are from the following ESC clones: Cluster miR-106a~363 on the X chromosome (A) Wild type JM8.A3, (B,C) miR-106a~363 null with selection cassette, (D) miR-106a~363 conditional, (E,F) miR-106a~363 null without selection cassette..
