The present invention discloses and claims a method of A. tumefaciens-mediated genetic transformation for Jatropha curcas characterized in that using cotyledons from Jatropha curcas mature seeds after cutting off 1/3-1/4 of the cotyledons base as recipients infected by A. tumefaciens， co-cultivating the cotyledon explants with A. tumefaciens， then transferring onto the medium free of selective agent to induce callus for 4 weeks， subculturing onto the selection medium with the kanamycin as selective agent to select resistant shoots， then subculturing the resistant shoots to obtain the resistant plants， and verifying the transgenic Jatropha curcas plants by molecular detection assay. Provided is an Agrobacterium tumefaciens-mediated gene transformation method for Jatropha curcas， comprising the step of using cotyledons from Jatropha curcas seeds after cutting off 1/3-1/4 of the base of the cotyledons as explants for infecting with Agrobacterium tumefaciens， co-cultivating the cotyledon explants with Agrobacterium tumefaciens， then transfering onto the medium free of selective agent to induce callus for 4 weeks， subculturing onto the selection medium with the selective agent of kanamycin to select resistant shoots， subculturing the resistant shoots to obtain the resistant plants， and verifying the transgenic plants by molecular detection assay.
