The goal of this project was to determine the differential expression of mRNA between two subtypes of Gbx2+ amacrine cells. These data will aid in distinguishing these two related, but distinct, amacrine cell subtypes by providing unique gene markers and physiological pathways. Overall design: Two amacrine cell subtypes were labeled in mouse retina with fluorescent tags, tdtomato and GFP using Gbx2-CreERT2-IRES-EGFP; Ai9 transgenic mice. Mice retinae from P6-7 mice were dissected and Tdtomato labeled amacrine cells were purified by flow cytometry. Tdtomato-labeled amacrine cells were separated into two groups, GFP-negative (Cell type 1) and GFP-positive (Cell type 2), by flow cytometry. 4 biological replicates, originating from 3 different mouse litters, were collected. RNA was extracted from 750-2500 sorted cells. One GFP and one Tdtomato cell type sample will be collected from each animal. There was at least 1 male and 1 female sample used in this study.
