This project aims to assemble the genome of the Bu Tuo Black Sheep (BBS), an important breed known for its high-altitude adaptability and disease resistance, using HiFi sequencing data. HiFi reads, which offer long read lengths and high accuracy, are crucial for generating high-quality genome assemblies. We have employed the hifiasm software, recognized as one of the best tools for assembling eukaryotic genomes from HiFi data. The hifiasm software, developed by Li Heng's team at Harvard University, is designed to handle PacBio HiFi data and produce phased assembly graphs that represent haplotype information, resulting in better resolution of segmental duplications and longer contigs. Our assembly approach leverages the speed and accuracy of hifiasm to generate a comprehensive and contiguous genome assembly for the BBS, which will provide valuable insights into its genetic makeup and contribute to breeding and conservation efforts. This study not only enhances our understanding of the BBS's genetic diversity but also paves the way for future genetic studies and applications in livestock genetics.
