The goals of this study are to explore the function of Pten in regulating the pluripotency transition of mouse embryonic stem cells through Pten knockout and mutation. Overall design: we generated Pten−/− and Pten-A3 mutant mouse ESCs with the CRISPR-Cas9 system and explored the effects on the pluripotency transition of mouse embryonic stem cells. We used a series of biological technologies such as Western blot, quantitative real time PCR, and cell culture to verity the conclusions.
