More and more steatotic livers undergo resection or transplantation but they exhibit higher susceptibility to ischemia-reperfusion injury (IRI), which results in increased perioperative complication morbidity and mortality. To investigate the underlying mechanisms, we conducted RNA-seq with liver samples derived from mice that were fed either high-fat-diet (HFD) or normal diet (ND) and experienced either IRI or sham operation. Overall design: Mice were fed either HFD or ND for 4 months. After liver steatosis model was constructed, the mice were subjected to either IRI or sham operation. For IRI, the left and middle branch of hepatic pedicle was ligated; after 1 h ischemia, the ligation was removed and liver reperfusion was confirmed visually; after 6 h reperfusion, the mice were anesthetized and liver samples were collected. For sham operation, the branch of hepatic pedicle was not ligated and the other procedures were the same as IRI. Thus, the experiment involves four groups: ND-Sham (NS), ND-IRI (NI), HFD-Sham (HS), HFD-IRI (HI).
