T cell gene repertoires in patients with Multiple Myeloma under treatment with daratumumab monotherapy

The presented evidence was acquired in the context of a prospective, multicenter, non-comparative, open-label, phase II study (ClinicalTrials.gov identifier: NCT03475628, accessed on 25 April 2022), evaluating effects of daratumumab monotherapy on bone parameters in patients with relapsed/refractory MM (RRMM) with at least two prior lines of therapy, including a proteasome inhibitor and lenalidomide (Celgene Corporation, Summit, NJ, USA), according to the first approved drug label in Greece.The main focus of the present analysis, as a secondary endpoint of this phase II study, was to evaluate the immune effects of daratumumab monotherapy on T cells of patients. In the present study we employed next generation sequencing (NGS) for TR gene repertoire profiling to comprehensively explore T-cell dynamics under daratumumab monotherapy in MM.
