To examine the transcriptional landscape of breast cancer stem cells enriched population in triple negative breast cancer, we compared transcript change between adherent MDA-MB-231 cells and the same cells cultured in mammosphere conditions for 16 hours or 5 days. Mammosphere assay is a recognised in vitro model of the breast cancer stem cells , and we wished to identify transcriptional dynamics and novel noncoding RNAs implicated in the regulation of this cell type. To define the role of identified player in mammospheres we next performed RNAi in these cells followed by RNA-seq Overall design: MDA-MB-231, in adherent, 16h sphere, and 5d sphere conditions, in duplicate. RNAi of crucial player in mammosphere formation.
