dCas9KRAB and gRNA targeting HERVK TEs were used to repress the TEs in stem cells (hiPS) and in differentiated induced neurons (iN). As control, dCas9KRAB alone without gRNA was used. hiPS cells have been transduced at an MOI15 with lentivectors coding for dCas9KRAB and gRNA targeting LTR5hs, or no gRNA as control. Transduced cells have been selected with puromycin and then induced to differentiate in neurons with doxycyclin (1µg/µl)-dependent expression of Neurogenins.
