Knowledge of genes/mechanism underlying heat stress tolerance in lentil can help to develop the heat tolerant lentil cultivars. In recent years, transcriptome sequencing has been used widely to identify candidate genes. In lentil, heat stress often affects the flowering and podding during reproductive stage. Therefore, candidate genes expressed under heat stress in the crop field would be more useful in the development of heat tolerant lentil cultivars. Earlier heat tolerant (IG4258) and heat sensitive (IG3973) lentil genotypes were identified and characterized on the basis of agrophysiological traits. These genotypes were also confirmed for their heat tolerance and sensitivity based on physiological and biochemical traits in other studies. These were used in the present investigation for RNAseq analysis. This analysis was conducted on leaf samples collected at normal and high temperatures from these genotypes. In the present study, highly tolerant (IG4258) and sensitive (IG3973) genotypes were used for identification and validation of the genes expressed at reproductive stage under heat stress in the field condition in lentil using RNAseq or transcriptome analysis.
