Profiling the inflammatory signatures of bulk MC38, Pan02 and KPC tumors from wild-type mice and Sirpα-KO mice before and after radiation therapy

The inflammatory gene expression profile in MC38, Pan02 and KPC tumors from wild-type mice and Sirpα-KO mice before and after radiation was analyzed by nCounter Analysis System using nCounter Mouse Immunology kit including 561 immunology-related mouse genes . MC38, Pan02 or KPC tumors (sizes 250-300mm3) in wild-type mice and Sirpα-KO mice were treated with a fraction of 8Gy X-ray radiation. Tumors were resected 12h and 18h post irradiation (n=2 mice per time point), followed by isolation of total RNA. RNA samples of the two time points were pooled and analyzed with Nanostring nCounter assays. Therefore, each set of Nanostring data represents transcription profile of four RNA samples (four mice).
