Analysis of the functional significance of peptide-N(4)-(N-acetyl-beta-glucosaminyl)asparagine amidase in human melanoma cells. The hypothesis tested in the present study was that this glycosidase may be involved in melanoma development. Overall design: Short hairpin RNAs were used to knockdown the expression of peptide-N(4)-(N-acetyl-beta-glucosaminyl)-asparagine amidase in human melanoma cells. The total RNA sample were isolated from the cell samples and prepared for global gene expression profiling using HT-12v4 Human Gene Expression Bead Chips and a HiScan array scanning system. The study included 12 experimental conditions. Each experimental condition had two biological replicates, for a total of 24 samples in the analysis. Samples with non-targeting shRNA (NT-shRNA) served as controls.
