In this experiment the relationship between SOX2 levels and WNT-induced gene expression was assayed using RNA-seq during embryonic stem cell differentiation. A doxycycline inducible cell line (SOX2 TetON) was employed to experimentally control SOX2 expression. Transcriptional profiles were generated from three independent biological replicates in four experimental conditions. Sox2 TetON ES cells differentiated in the presence or absence of 1ug/ml doxycycline were compared to differentiated WT epiblast progenitor cells cultured in the presence or absence of the WNT agonist CHIR99021.
