Mesenchymal stromal cells (MSCs) are a phenotypically diverse cell type that exhibit numerous therapeutic properties including regenerative traits, immunomodulation and antibacterial activity. However, MSCs are notoriously heterogenous and exhibit source-dependent phenotypes. In this study, bone marrow-derived MSCs from two mouse strains (C57BL/6 and BALB/c) were examined for their ability to inhibit bacterial growth. This project data set includes population-level RNA seq for MSCs during routine growth, and after exposure to the bacterial lipopolysaccharide (LPS). In this work, we identified the upregulation of the LPS co-receptor CD14 enhanced LPS recognition kinetics and improved antibacterial properties in these MSCs. This data set also includes single-cell RNA-seq using the 10x Genomics platform of CRISPRa-mediated CD14 overexpressing MSCs in response to LPS stimulation.
