Pre-mRNA fate decision by the RBP TTP. TTP-bound RNA target analysis in RAW264.7 macrophages (murine) upon LPS stimulation.

Total RNA sequencing analysis of tristetraprolin (TTP; gene name Zfp36) bound RNA targets in murine RAW264.7 macrophages. In TTP KO RAW264.7 macrophages, a doxycycline inducible 3x C-terminally Myc-tagged TTP was expressed. Subsequently freshly transcribed RNAs in response to 6 h LPS stimulation were metabolically labeled with 4sU. After gently crosslinking, cells were lysed and enriched for TTP. TTP bound RNAs were released by proteinase K digest and together with total cell RNA extracts (input) submitted for RNA seq analysis.
