Since oxidative stress and neutrophil infiltration play a central role in ischemia-reperfusion injury (IRI), the effects of epigallocatechin gallate (EGCG) on neutrophil infiltration-related markers were investigated. The IRI model utilizing an elastic rubber band was 4 + 2 h (IRI group). In sham-control (SC) group, all steps but band binding were preserved. Saline (10 mL/kg/i.p.) and EGCG (25 and 50 mg/kg/i.p.) were administered 30 minutes before the reperfusion (IRI-SF, IRI-25, and IRI-50 groups). Creatine phosphokinase (CPK), lactate dehydrogenase (LDH), total oxidant status (TOS), total antioxidant status (TAS), myeloperoxidase (MPO), E-selectin, P-selectin, L-selectin, intercellular adhesion molecule-1 (ICAM-1), and various cytokines (IL-1β, IL-6, TNF-) were determined in serum and gastrocnemius muscle samples. CPK, LDH, and TOS levels were higher in the IRI group compared to the SC group; insignificant decreases were observed in the IRI-50 group compared to the IRI group. Regarding TAS, a visible decrease was seen in the IRI compared to SC group, which was significantly increased by 50 mg/kg of EGCG. Regarding remaining parameters, statistically insignificant increases in IRI compared to SC and decreases in the EGCG-treated versus IRI groups were detected. We claim that EGCG might have a beneficial effect via an interaction with neutrophil infiltration-related markers.
鉴于氧化应激与中性粒细胞浸润在缺血再灌注损伤(ischemia-reperfusion injury, IRI)中发挥核心作用,本研究探讨了表没食子儿茶素没食子酸酯(epigallocatechin gallate, EGCG)对中性粒细胞浸润相关标志物的影响。本研究采用弹性橡皮筋构建时长为4+2小时的缺血再灌注模型,该组设为IRI组。假手术对照组(sham-control, SC)仅省略橡皮筋结扎操作,其余实验步骤均与IRI组保持一致。于再灌注前30分钟,分别向各组给予腹腔注射生理盐水(10 mL/kg,对应IRI-SF组)、25 mg/kg EGCG(对应IRI-25组)及50 mg/kg EGCG(对应IRI-50组)。采集血清及腓肠肌样本,检测肌酸激酶(creatine phosphokinase, CPK)、乳酸脱氢酶(lactate dehydrogenase, LDH)、总氧化状态(total oxidant status, TOS)、总抗氧化状态(total antioxidant status, TAS)、髓过氧化物酶(myeloperoxidase, MPO)、E-选择素、P-选择素、L-选择素、细胞间黏附分子-1(intercellular adhesion molecule-1, ICAM-1)及多种细胞因子(IL-1β、IL-6、TNF-α)的水平。与SC组相比,IRI组的CPK、LDH及TOS水平显著升高;相较于IRI组,IRI-50组上述指标仅出现无统计学意义的下降。就TAS而言,IRI组较SC组出现明显降低,而50 mg/kg EGCG可显著逆转这一降低趋势。对于其余检测指标,IRI组较SC组仅存在无统计学意义的升高,EGCG干预组相较于IRI组则呈现无统计学意义的下降趋势。本研究表明,EGCG或可通过调控中性粒细胞浸润相关标志物发挥有益的保护作用。
