We investigated the impact of several experimental conditions on the methylation level of a single individual following custom targeted genome bisulfite sequencing. Impacts of DNA fragmentation, input DNA quantity, probe dilution and the number of PCR cycles were quantified by comparing the distribution of methylation over 18Kb genomic sequence in the same individual.
