Background: Rheumatoid arthritis (RA) involves complex immunological and metabolic dysregulation, suggesting that multi-targeted therapeutic strategies may offer advantages over conventional single-target approaches. Huangqi Guizhi Wuwu Decoction (HGWD), a classical Traditional Chinese Medicine formula with established clinical use in RA, represents a promising source of multi-targeted natural products; however, its bioactive constituents and mechanisms require systematic investigation. Methods: This study integrated serum pharmacochemistry, network pharmacology, untargeted metabolomics, and experimental validation to comprehensively characterize the therapeutic mechanisms pharmacodynamic basis of HGWD. Conclusions: These findings provide mechanistic insights into HGWD's multi-component therapeutic effects and identify promising bioactive constituents for the development of multi-targeted natural product-based RA therapeutics.The data used in this study are authentic and reliable, and have been approved by the ethics review.
