The goal of this experiment is to identify genes differentially expressed in MEF cell lines with a targeted MDMX mutation that prevents binding and inhibition of CK1-alpha kinase. Overall design: MDMX-SG (MDMX with W201S/W202G substitutions) knock-in mutant mouse was crossed into p53-null background to generate p53-/-;MDMXSG/SG homozygous MEFs (SG) and p53-/-;MDMXWT/WT control MEFs (WT). Three cell lines of each genotype (SG1/SG2/SG3 versus WT3/WT9/WT11), each cell line generated from one embryo, were compared to identify genes affected by the MDMX-SG mutation.
