The authors used a co-culture of primary human brain microvascular endothelial cells (HBMEC) with <i>Plasmodium falciparum</i> material, recombinant PfEMP1 or lysates from infected erythrocytes (IE), and measured barrier function by Trans Endothelial Electrical Resistance (TEER). A selection of Protease-Activated Receptor-1 (PAR1) inhibitors was tested for their ability to reverse the <i>P. falciparum</i> and thrombin induced decrease in barrier function.<br>The TEER data is from an initial screen with 10 PAR1 inhibitors and the human umbilical vein endothelial cell line EA.hy926 followed by more detailed testing of 4 selected inhibitors and HBMEC. In addition, the effect of IE or IE lysate on EPCR expression on HBMEC was determined by flow cytometry.
