Histone Methyltransferase G9a in Primary Sensory Neurons Promotes Inflammatory Pain and Transcription of Trpa1 and Trpv1 via Bivalent Histone Modifications

This RNA-seq experiment was aimed to investigate genome-wide differential gene expression by Ehmt2 (G9a) knockout in dorsal root ganglion (DRG) neurons in mice treated with complete Freund's adjuvant(CFA) or vehicle. Ehmt2 ablation significantly reversed expression of 981 genes in the DRGs and among these genes 322 were downregulated and 659 genes are upregulated (P<0.05). Total RNA was isolated from DRGs in Ehmt2 (G9a) inducible conditional knockout (Avil-iCreERT2/AJwo /G9aflox/flox, G9a-iKO) and littermate control (WT, without Cre) mice. Both WT and G9a iKO mice were treated with vehicle or CFA and 14 days after, RNA from four groups were sequenced on illumina NovaSeq 6000 platform. Each group has two repeats. Differential gene expression was analyzed using Hisat2-FeatureCounts-Deseq2 pipeline.
