Datasets for the publication "Unjamming transitions in cancer cell spheroids are linked to EMT-dependent cell-matrix interactions", to be submitted.Datasets include the raw tif files of 3D spheroid invasion assays of A549 and MV3 spheroids embedded in collagen (2.4 and 8 mg/mL) and gelMA (30 and 50 mg/mL). This include time-series bright-field imaging for 24 hrs (MV3) and 72 hrs (A549) at one hour interval. The spheroids were treated with exogenous TGF-β and broad spectrum MMP-inhibitor and imaged under same conditions.Datasets include MATLAB/.m codes to analyze the spheroid images for effective spheroid radius, number of disseminated cells, rate of invasion and protrusion length. Additionally, MATLAB codes/.m to plot figures are also included within each respective condition. Raw data for supplementary data is also provided as images, excel files and MATLAB scripts.
本数据集对应待投稿论文《肿瘤细胞球体解堵转变与上皮间质转化(Epithelial-Mesenchymal Transition, EMT)依赖的细胞-基质相互作用(Unjamming transitions in cancer cell spheroids are linked to EMT-dependent cell-matrix interactions)》。数据集包含A549与MV3细胞球体的三维球体侵袭实验原始TIF文件,这些球体分别包埋于浓度为2.4 mg/mL与8 mg/mL的胶原蛋白及30 mg/mL与50 mg/mL的gelMA中。实验采用明场时序成像:针对MV3细胞球体采集24小时的时序图像,针对A549细胞球体采集72小时的时序图像,采样间隔为1小时。所有细胞球体均经外源性转化生长因子-β(Transforming Growth Factor-β, TGF-β)及广谱基质金属蛋白酶(Matrix Metalloproteinase, MMP)抑制剂处理,并在相同实验条件下完成成像。数据集附带用于分析球体图像的MATLAB/.m代码,可计算有效球体半径、扩散细胞数量、侵袭速率及伪足长度;此外,各实验条件下均配有用于绘图的MATLAB/.m代码。补充数据的原始资料以图像文件、Excel表格及MATLAB脚本的形式提供。
