Although the microbiota in prostatic tissue of patients with prostate cancer have been studied, results of microbiota in each study are conflict. Potential contaminations during sampling collection and experiments could influence on microbiome results of prostatic tissue as low-biomass sample as well as difference of ethnic and study design. In this study, we analyzed the microbiota and their potential functions in benign and malignant tissue of prostate cancer with considering possible contaminants and host variables. We analyzed microbiota from 118 tissue samples (59 benign tissue and 59 malignant tissue) obtained by robot‐assisted laparoscopic radical prostatectomy and 64 negative controls (from sampling to sequencing processes). The alteration of microbiome in prostate tissues was detected only in patients with diabetes, and the influence of diabetes on the microbiome was significant in malignant tissue. The microbiome in malignant tissues of patients with diabetes was influenced by pathologic stages. The relative abundance of Cutibacterium was reduced in high pathologic group compared to intermediate group, and this reduction was related to the increased microbial pathways in high pathologic group. Results in this study indicate that diabetes should be considered as important host covariate in microbiota study of prostatic tissue. Although further studies are necessary to evaluate findings in this study, this study can help to understand tissue microbiome in prostate cancer.
