In order to investigate the response of epithelium to butyrate, we generated in vitro epithelial organoid cultures from colon samples of non-IBD controls and they were stimulated with different doses of butyrate. The transcriptional signature revealed that butyrate negatively regulated proliferation and cell cycle, induced a protective response to oxidative stress and regulated genes related to the immune response. Colon samples from non-IBD controls (subjects undergoing surgery for colorectal cancer) were collected. Epithelial organoid cultures (EpOCs) were expanded from human colonic crypts and then induced to differentiate (d-EpOCs). Paired first-passage EpOCs and d-EpOCs (n=4) were stimulated with two doses of butyrate (0.5 mM and 5 mM) during 1h, 3h, 6h and 24h. RNA was then extracted and used for hybridization on Affymetrix microarrays.
