This study aimed to elucidate the molecular mechanisms underlying TMJ-OA by integrating subcellular spatial transcriptomics with single-cell RNA sequencing analysis in TMJ-OA mouse models. Overall design: The TMJ-OA model mouse experiments were conducted using 9-week-old female C57Black/6J mice. The TMJ-OA models were divided into three groups: Control (n=7), Mechanical Stress (MS) model (n=7), and Articular Disc Displacement (ADD) model (n=4). scRNA-seq was performed on cells obtained from the articular disc, including the synovium, three weeks after establishing the TMJ models.
