ONT nanopore sequencing allows direct sequencing of RNA molecules. But these is a lack of formal barcoding protocol for direct RNA sequencing. In this experiment, we developed barcoding and demultiplexing strategy. To avoid nanopore's high basecalling error rate, we developed a deep convolution neural network classifier that works on the raw signal associated with the DNA barcode used for barcoding RNA molecules. Therefore, we manage to offer a simple and cost-effective barcoding and demultiplexing strategy for direct RNA sequencing, which will enhance the use of nanopore technology.
