Purpose: to identify the effects and kinetics of a panel of polarizing cytokines on Th17 differentiation in vitro Naïve CD4 T cells were isolated from C57BL/6 mice and the cells were sort-purified and cultured in the presence of plate-bound αCD3 and plate-bound αCD28. Th17 differentiation was induced by culturing the cells in the presence of TGFβ , IL6 , IL21 and/or IL1β . Part of the cells was cultured without any polarizing cytokines, in “Th0” conditions. Three biological replicates of samples were collected at 0, 0.5, 1, 2, 4, 6, 12, 24, 48, and 72 hours time points.
