Immuno-oncology NGS panel, a new tool for investigating mouse tumor immunity

Immunogenomics has become critical in investigating tumor immunology, mechanisms of actions and biomarkers of immunotherapy. Mouse tumor modeling, the foundation for immuno-oncology (IO) research, can be more productive if equipped with immunogenomics tools. A new 1080-mouse-IO RNAseq panel (NGSmIO) was compared with microarray or whole transcriptome sequence (RNAseq) for assessing mouse tumor models, showing good correlations. However, NGSmIO has broader dynamic range than microarray assay, particularly for rare transcripts, and better correlation with RNAseq than microarray. NGSmIO is more sensitive (>10x deeper) than RNAseq per unit data. The lineage-identification gene signatures included in NGSmIO reveals tumor-infiltrate leukocyte components, correlating with that of FACS, implicating lineage detection/quantification utility. NGSmIO-panel can be a robust, faster and cost-effective solution for preclinical immuno-oncology research.
