Altered lipid metabolism contributes to lung inflammation and drives lung tumorigenesis. A key factor maintaining homeostasis is Sphingosine Kinase 1 (SPHK1), which phosphorylates sphingosine to the bioactive lipid sphingosine-1-phosphate. Interruption of this synthesis pathway in lung tumor cells promotes a pro-tumorigenic microenvironment and hence drives tumor progression. To better understand this process, we performed RNAseq analysis of SPHK1 proficient and deficient mouse lung tumor cells harboring KrasG12D mutations. We established KrasG12D driven, p53 deficient lung tumor cell lines. Subsequently, we knocked out SPHK! In these cells. For RNAseq analysis, we harvested RNA before and after treatment witth 10% fetal calf serum.
