miRNA sequencing of exosome derived from endothelial progenitor cells in Rattus norvegicus

Bone marrow-derived endothelial progenitor cells were isolated and cultured from 4-week-old rats (Rattus norvegicus), then divided into two groups: Control group (CON group) and Shock wave group (SW group). SW group was intervened by extracorporeal cardiac shock waves (ECSW), but the CON group was conducted without ECSW. The cells were then cultured for 48 hours until cell culture media was collected for harvesting exosomes. For SW-exo and CON-exo(n=4), Next generation sequencing was performed by LC Sciences (USA) in accordance with the standard procedures provided by Illumina, including miRNA sequencing library preparation, sequencing process, and NGS data analysis. Based miRNAs sequencing, the results revealed that differentially expressed miRNAs delivered by exosomes may exert pivotal effects in extracorporeal cardiac shock waves treatment.
