Breeding and re-introduction of endangered firefly Pyrocoelia pectoralis to habitat becomes necessary. An improved chromosome-level genome assembly of P pectoralis was conducted and SSR markers were developed to confirm sperm competition and paternity. The assembled genome size is 532.25Mb. Hi-C analysis assembled scaffolds onto 10 chromosomes and a scaffold N50 of 52.87Mb, with more than 96% of assembled bases located on chromosomes. This is the first report to study sperm competition and paternity by SSR in fireflies. In total, 13 SSR markers were developed. Both anatomy of female reproductive system and paternity experiments confirm sperm competition in P. pectoralis. Understanding Sperm competition and paternity study based on chromosome level genome assembly help conservation of endangered firefly P. pectoralis.
