Effect of SARS-CoV-2 mutants on human DC maturation

SARS-CoV-2 mutants have different effect on maturation of monocyte-derived DC (Mo-DC): Representative histograms show the flow cytometry analysis for DC maturation. Briefly, Mo-DC were infected with SARS-CoV2 or its mutants (MOI=3) or LPS. After incubation (1-3 days), cells were stained with antibodies for CD11c, CD80, CD86 and MHCII for 10 min at room temp, washed once with PBS, fixed twice with 4% PFA, removed from BSL3 and analyzed in LSRFortessa. The % CD11c+ cells expressing CD80, CD86 or MHCII, and MFI for each marker were calculated. The MFI (median) of each marker expression was also calculated. The histograms presented in this publication show the gating strategy for CD80 expression in DC.
