We analyzed the transcriptomic profile of peritoneal macrophages (PMs) directly sorted from two groups of mice fed a Western diet for 16 weeks: (1) mice with LysMCre-mediated myeliod-specific Lipa overexpression on the Ldlr-/- background; (2) littermates that are LysMCre-negative and are on the Ldlr-/- background. Overall design: We generated mice for Lipa overexpression (LipaTg/wt) on C57BL/6 background by inserting Lipa open reading frame (NM_001111100) into the Rosa26 locus. To achieve myeloid-specific Lipa overexpression, we bred LipaTg/Tg, Ldlr-/- mice with LysMCre+/-, Ldlr-/- mice. The obtained LysMCre+/-, LipaTg/wt, Ldlr-/- (LipaTg) and LysMCre-/-, LipaTg/wt, Ldlr-/- (Ctl) littermates were fed a Western diet for 16 weeks. The LipaTg mice demonstrate myeloid-specific Lipa overexpression, where eGFP acts as a marker for cells with successful cre-loxp recombination leading to subsequent Lipa overexpression. The viable CD45+F4/80+ PMs from Ctl mice and CD45+F4/80+eGFP+ PMs from LipaTg mice were sorted and directly subjected to RNA-seq without culturing. n = 3 female mice per genotype.
