Mechanical force regulates various biological responses in many cell types. Here, the retinoic acid treated mouse gingival fibroblast derived induced pluripotent stem cells were treated with intermittent compressive for 24 h and gene expression profiling was evaluated using RNA sequencing technique. Results indicated that intermittent compressive stress regulated various pathways in these cells including p53 signaling and other metabolic pathways. Mouse gingival fibroblast derived induced pluripotent stem cells were cultured in low attachment plate to form embryoid bodies and subsequently treated with retinoic acid. Retinoic acid treated embryoid bodies were then transfer to gelatin coated dishes. The intermittent compressive stress was treated in serum free culture condition with 1.5 g/cm2 with the loading frequency of 0.23 Hz. Total RNA was collected at 24 h after loading and prepared for gene expression analysis using RNA sequencing technique.
