Effect of spatial effect on hESC cell fate determination under BMP4 induction

hESC cell differentiation under BMP4 induction have spatial effect. in hESC colonies, cells could be stained by two populations by JC-1 staining, the JC-1 positive cells in outlayer of colonies and the JC-1 negative cells in the central of colonies. We also found that only JC-1 positive could be differentiated under BMP4 induction, so we performed the sequencing by JC-1 sorting method. in order to study the conditions that promote BMP4 induced cell differentiation, we also treated cells by different chemicals under BMP4 induced cell differentition.
