This study aims to explore the chromatin accessibility landscape in peripheral blood mononuclear cells (PBMCs) of centenarians to understand the epigenetic basis of exceptional human longevity. We performed ATAC-seq on PBMC samples collected from centenarians and age-matched elderly controls. By profiling genome-wide open chromatin regions, we identified a distinctive chromatin accessibility signature in centenarians, characterized by increased accessibility at regulatory elements, particularly in B cells. Integration with transcriptome data revealed transcriptional reprogramming associated with chromatin remodeling. These findings provide novel insights into the epigenetic regulation of immune aging and highlight potential mechanisms contributing to healthy longevity.
