This study aims to systematically analyze the community structure and functional characteristics of the oral microbiome in children with dental caries using metagenomic sequencing technology. Furthermore, it investigates the effects of two active compounds, hesperidin and naringin, on microbial gene expression and functional regulation in the oral cavity of children with dental caries, providing theoretical foundations for caries prevention and the application of natural active compounds. A total of 11 saliva samples from children with dental caries were collected, with each sample divided into three groups: a blank control group (no intervention), a hesperidin experimental group (intervention with hesperidin), and a naringin experimental group (intervention with naringin). Total DNA was extracted from the processed saliva samples in all three groups, followed by metagenomic sequencing to obtain microbial genomic data. Bioinformatics analysis was conducted to compare the differences in microbial gene abundance between the blank control group and the two experimental groups, thereby evaluating the regulatory efficacy of hesperidin and naringin on the gene abundance of the oral microbiome in children with dental caries. The results revealed significant differences in gene abundance composition between the control and experimental groups, with both hesperidin and naringin demonstrating regulatory efficacies exceeding 70% on the gene abundance of oral microorganisms in children with dental caries, indicating that these two active compounds exert substantial intervention effects at the genetic level of the oral microbiome.
