Transcriptional changes of tumoral dendritic cells with tumor progession

Dendritic cells (DCs) play important role of orchestrating anti-tumor immune responses both in tumor context. Following tumor progession, DCs were dysfunctioned by chaotic tumor microenvironment. To understand the change in the molecular level, we purfied conventional DCs from tumor at early stage (day 7 and around 200mm3) or late stage (day14 and larger than 600mm3) and sent for sequencing. Additonally, we would like to understand how pde5 deletion affects DCs function. Tumoral DCs were sorted from MC38-bearing Zbtb46-Tdtomato mice at early- (day7) or late-stage (day14) for seqencing. BMDCs derived from WT or Pde5-/- mice were stimulated with or without 200 ng/ml LPS for 12h and then collected for sequencing.
