The innate immune system plays key roles in tissue regeneration. For example, microglia promote neurogenesis in Müller glia in birds and fish after injury. Although mammalian retina does not normally regenerate, neurogenesis can be induced in mouse Müller glia by Ascl1, a proneural transcription factor. We show that in mice, microglia inhibit the Ascl1-mediated retinal regeneration, suggesting the innate immune system limits the regenerative response to injury. Overall design: Five samples were created for the publication Microglia suppress Ascl1-induced retinal regeneration in mice. One sample contains Cd45+/Cd11b+ cells from ANT treated mice seven days after injury. These contain immune cells in the mouse retina. The remaining four samples are ANT reprogrammed FAC-sorted Muller Glia at 2 days and 7 days post ANT treatment with and without the CSF1R inhibitor PLX5622.
