We produced the transgenic mice expressing antisense-Dnmt3L under the Miwi2 promoter (Tg-Miwi2P-asDnmt3L).Tg-Miwi2P-asDnmt3L mice showed reduced endogenous Dnmt3L RNA expression and a phenotype quite similar to that of DNMT3L-difficient mice. We examined the genomic insertion sites of transgene to clarify the mechanism of piRNA-mediated silencing.
