We performed ChIP-seq analysis in order to determine the transcriptional targets of p53 in the CML cell line BV173 as a result of p53 activation using the MDM2 inhibitor idasanutlin. We cultured one bioreplicate of 1 x 10^7 BV173 cells with and without idasanutlin (300 nM) for 6 hrs and then performed ChIP using a ChIP-grade antibody for p53. We isolated ChIP DNA from the p53 ChIP conditions. In parallel, we treated one bioreplicate of BV173 cells with and without idasanutlin (300 nM) and prepared input genomic DNA (control) from cross-linked cells. In total, we had four experimental conditions.
