This article investigates the impact of the first five codons on translation efficiency in Escherichia coli. By constructing a GFP library containing nearly all possible nucleotide combinations at the 3rd to 5th codon positions, researchers found that certain combinations significantly enhanced GFP expression, although this effect was minimal in endogenous genes. These nucleotide combinations were also enriched in other enterobacteria, suggesting evolutionary pressure to select these sequences for improved translation efficiency, and their potential applications in biotechnology were explored.
