Biochemical fractionation of HL-1 mouse cardiomyocyte nuclei and RNA-seq of chromatin-enriched and soluble-nuclear RNA Overall design: Three biological replicates of HL-1 cell nuclei were subjected to biochemical fractionation. ERCC spike-in RNA standards were added to each fraction prior to RNA extraction. RNA from three chromatin pellet and three soluble nuclear fractions were converted to cDNA using NEBNext Ultra II Directional RNA library peparation and sequenced on the Illumina HiSeq4000.
