Characterization of naive CD4 T cells deficient for Foxo1 expression in T cells

Naive CD4 T cells lacking Foxo1 expression exhibit an altered transcriptional signature. CD4 T cells from peripheral lymph-nodes of mice deficient (C57BL/6-Foxp3GFP Foxo1TKO) or proficient (C57BL/6-Foxp3GFP Foxo1Ctrl) for Foxo1 expression in T cells were first purified by depleting B cells and CD8 T cells. Then, Naive CD4 T cells were FACS-sorted as CD44-/low GFP- Lin- cells and mRNAs extracted.
