Gone with the Wind: High Wind Speeds Reduce Targeted Species Detection with Airborne eDNA

Raw data files, analysis scripts, and input files to generate data presented in the manuscript.\nLineage: DNA was collected from the air using Modified Wilson and Cooke air samplers. These were then converted into water samples by adding 100ml of deionised water.\nWe then pumped this water through a 3 µM polycarbonate isopore size filter 25 mm in diameter. We then used a Qiagen DNeasy Plant Mini Kit to extract the DNA from these filters.\nDNA fragments were then amplified on a QuantStudio 3 using the primer pairs:\ntrnS-trnG\nForward primer (trnS-trnG) 5'-TCACCCCGTTTTTCAAATTTCA-3' \nReverse primer (trnS-trnG) 5'-ACCACTAAACTATACCCGCTAC-3'\nITS1\nForward primer (ITS1) 5’-GGGAGGGGATGACCACGG-3’ \nReverse primer (ITS1) 5’-TGCAAGAGCCGAGATATCCG-3’\nThe resulting ITS1 sequences were then analysed using sanger sequencing.\nThe sequencing reactions were carried out using the BigDye Terminator v3.1 Cycle Sequencing Kit, following the manufacturer's protocol. The reactions were analysed on an ABI 3730xl DNA Analyzer.
