The effects of temperature and moisture on nitrogen mineralisation in wetland soils of varying degrees of degradation were determined using an in vitro cultivation method. Three temperature levels (15, 25, 35 °C) were established. Based on soil moisture content variations during wetland degradation, four soil moisture levels were set: 20% field capacity (20% FC), 40% field capacity (40% FC), 60% field capacity (60% FC), and 80% field capacity (80% FC). A full factorial experimental design was employed for temperature and moisture, yielding 12 temperature-moisture combination treatments. Soil samples were collected from four wetland degradation levels (undegraded, slightly degraded, moderately degraded, severely degraded) under each temperature and moisture condition, yielding 48 treatment combinations. Each treatment was replicated three times, resulting in 144 soil samples. These samples were incubated in a constant-temperature incubator. Changes in soil nitrogen mineralisation were measured over 3–49 days.
